CBN (cannabinol; molecular formula C21H26O2) is a degradation product rather than a primary biosynthetic compound. As delta-9-tetrahydrocannabinol (THC) is exposed to light, heat, and oxygen over time, the compound undergoes oxidative degradation and the side chain cyclises, yielding CBN. Fresh cannabis material contains relatively little CBN; concentrations rise measurably in improperly stored or aged plant material. It was, historically, one of the first cannabinoids isolated and characterised , identified in the late nineteenth century before THC's structure was even known.
At the receptor level, CBN acts as a partial agonist at both CB1 and CB2 cannabinoid receptors, though with substantially lower binding affinity at CB1 than THC. This reduced CB1 affinity corresponds to markedly weaker psychoactivity. CBN also shows affinity for transient receptor potential (TRP) channels, including TRPA1 and TRPV2, and has been reported in preclinical studies to interact with TRPM8. The functional significance of these non-CB receptor interactions in humans remains under active investigation.
Preclinical research has examined CBN's pharmacological profile across a range of endpoints , including effects on immune-cell function via CB2 receptors and its behaviour as a weak agonist in rodent bioassays , but the volume of rigorous human clinical trial data is limited. Researchers have noted the practical difficulty in isolating CBN's effects from those of co-occurring cannabinoids and terpenes, particularly in whole-plant preparations. The existing evidence base is not sufficient to support conclusions about clinical utility, and no CBN-specific therapeutic indication is currently approved by the Therapeutic Goods Administration (TGA) in Australia.
In Australia, CBN derived from cannabis is regulated under the Poisons Standard (the Standard for the Uniform Scheduling of Medicines and Poisons). Depending on concentration and formulation, cannabis-derived CBN preparations may fall under Schedule 4 (prescription only) or Schedule 8 (controlled drug) of that Standard. Any product containing CBN and intended for human therapeutic use must hold TGA registration or approval unless an exemption applies.
