Caryophyllene Oxide

Caryophyllene oxide is the oxidised derivative of beta-caryophyllene, a bicyclic sesquiterpene epoxide found widely across flowering plants and notable for its distinctive dry, woody, spiced aroma.

Caryophyllene oxide (molecular formula C15H24O) is a naturally occurring sesquiterpene epoxide produced when beta-caryophyllene undergoes oxidation, either enzymatically within plant tissue or through exposure to air post-harvest. It carries a dry, woody, clove-like scent with faintly waxy, herbal undertones , a profile that distinguishes it from its parent compound. It is the constituent responsible for cannabis being detected by drug-detection dogs, a property documented in forensic literature owing to its relatively high vapour pressure and persistence.

In nature, caryophyllene oxide occurs across a broad range of botanical families. It is found in the essential oils of black pepper (Piper nigrum), clove (Syzygium aromaticum), rosemary (Salvia rosmarinus), hops (Humulus lupulus), and various Eucalyptus species native to Australia, among many others. Its presence in a plant's volatile fraction is typically quantified by gas chromatography–mass spectrometry (GC-MS).

Preliminary laboratory research has examined caryophyllene oxide's interaction with biological systems at the cellular and molecular level. Like beta-caryophyllene, it has attracted interest for its relationship with the endocannabinoid system; however, existing data are largely derived from in vitro and animal models. No conclusions about effects in humans can be drawn from this body of work, and no therapeutic applications have been established. Researchers continue to characterise its chemistry and occurrence as a step toward understanding the full volatile profile of complex botanical extracts.

In the fragrance and flavour industries, caryophyllene oxide is used as a reference standard and as an aroma compound in its own right, valued for adding depth to woody and spice-forward compositions. Its status as a naturally derived compound means it appears on several international flavour ingredient registers, though regulatory classification varies by jurisdiction and intended use.
